疟原虫基因转染的研究进展 3

对该基因确切功能的阐述。另外,对于多拷贝基因的完全敲除非常困难。

  疟原虫转染技术最重要的应用就是基因敲除,但由于上述缺点迫切需要建立可诱导性转基因系统。目前认为四环素抑制子(tetracycline repressor,TetR)系统是最符合逻辑和最有前途的诱导性系统,该系统已在寄生原虫锥虫[35]和阿米巴[36]成功建立。其原理是调节基因表达的TetR与四环素抗性操纵子的调控序列结合,使RNA聚合酶不能与启动子结合,抑制下游结构基因的表达,当不同浓度的诱导物(四环素或其类似物)存在时,可不同程度地与TetR结合,使结合有四环素的TetR不能与调控序列结合,从而不同程度的开启结构基因的表达。

  此外,疟原虫基因转染的效率有待于进一步提高。
